Why Calorimetry?

Life is an endless chain of biochemical reactions, which favorability is determined by the thermodynamics of the system. This is why calorimetry is one of the most important techniques in diverse fields of basic and applied sciences, from Biophysics to Food and Pharmaceutical sciences. An advantage of calorimetry lies in its unique ability to access one of the thermodynamic state functions of a system, the enthalpy (ΔH). Since calorimetric measurements are done by detecting heat, and the heat is not a state function, it is important to make sure that the system is in equilibrium that is that the measured process is reversible. In practice this means that a denatured protein can refold at appropriate conditions and that a ligand can dissociate from a macromolecule when its population has been decreased.

The two most common techniques, Differential Scanning Calorimetry (DSC) and Isothermal Titration Calorimetry (ITC,) can be used interchangeably or can supplement each other. However, each of them has its own area of a most popular use:

  • DSC is used for studying stability of folding/unfolding transitions
  • ITC is applied for investigation of intermolecular interactions.
